Growth protocol MDA-MB-231 expressing control GFP or GATA3 were grown in DMEM/high glucose supplemented with 10% FBS, sodium pyruvate and pen/strep. 1.5 million cells were seeded into 100 mm dishes and RNA collected after 2 days.
Extracted molecule total RNA
Extraction protocol RNA was isolated by Trizol® according to the manufacturer's instructions. All samples used for microarray analysis had a high quality score (RIN >9).
Label biotin
Label protocol RNA (1 ug) was reverse transcribed with a T7-oligo(dT) primer and biotin labeled using the Affymetrix One Cycle Target Labeling kit (Affymetrix) following the manufacturer’s protocol.
 
Hybridization protocol Hybridized to Affymetrix human U133 plus 2.0 GeneChips
Scan protocol Scanned on an Affymetrix GeneChip scanner 3000 and data collected using Affymetrix GCOS software
