Treatment protocol |
Expression of ANGPTL4 was silenced in HUVECs (60-70% confluent, P4) by using specific sets of siRNAs using RNAiMAX according to manufactures instructions. 36h post trasfections cells were treated with either CoCl2 (100uM) or appropriate vehical control (PBS) for aditional 24h. In another sets of experiment, 36h posttrasfection, cells were treated either XEN445 (10uM) or apropriate vehical control (DMSO) for aditional 24h.

Growth protocol |
HUVECs (Human umbilical vein endothelial cells, P2) were obtained from the Yale School of Medicine, Vascular Biology and Therapeutics Core facility and cultured in M199/EBSS medium supplemented with 20% fetal bovine serum (FBS), 2 mM Glutamine and antibiotics in humidified atmosphere with 5% CO2 at 37C.

RNA was harvested using Rneasy mini plus kit (Qiagen). 1.3 ug of total RNA was used for the construction of sequencing libraries. RNA libraries were generated from control cells using TrueSeq Small RNA Library preparation (Illumina)
