Treatment protocol |
Cytobrushes (Cooper Surgical) were used to collect vaginal samples from ~5cm into the vagina. These were suspended in 700ul RNAlater (Ambion).

Extraction protocol |
TRIzol was used to extract total RNA from samples within 72 hours of collection following the manufacturer’s instructions. An extra step was added after homogenization and before addition of chloroform where excess un-homogenized material was cleared from the TRIzol by centrifugation at 12000g for 10 min.

Labeling was carried out with the Ambion WT Expression Kit for Affymetrix GeneChip Whole Transcript WT Expression Arrays and Affymetrix GeneChip WT Terminal Labeling kit as per the manufacturer's instructions.
