Extraction protocol Total RNA was isolated from adipose tissues using QIAzol Lysis Reagent as manufacturer's instruction. 5ug of total RNA was used for the construction of sequencing libraries.
RNA-Seq libraries were prepared with a TruSeq Stranded mRNA Library Prep kit (Illumina) using purified RNAs isolated from BAT of Ssu72 WT and aKO mice

Data processing Transcriptomic sequencing was performed on a Novaseq platform. An initial sequence-level quality assessment was performed using FastQC. RNA-Seq reads were then mapped to the human mm10 reference genome using Tophat (v2.0.13). Assembly, quantification and differential expression analysis were performed using Cuffdiff (v2.2.1)
