Total RNA was extracted from 1.5 x 107 CBMNC or APBMNC with RNeasy Kit (Qiagen). Quantity and purity of extracted RNA was checked by RNA LabChip® Kit (Agilent).

Biotinylated labeling cRNA was conducted by in vitro transcription using the BioArray High Yield RNA Transcript Labeling System (ENZO). cRNA was purified by GeneChip Sample Cleanup Module (Affymetrix).

Hybridization protocol |
Biotinylated cRNA was heated for 5 min at 95°C, allowing RNA to breakdown to small fragments according to Affymetrix protocols. Fragmented cRNA (20 µg) was subjected to oligonucleotide hybridization (Hybridization Oven; Affymetrix) to the human Genome Focus Genechip (Affymetrix).
