Growth protocol |
Peripheral blood was collected using PAXgene tubes, and then stored immediately to -80 degree freezer until ready for RNA extraction. Cells were not grown or cultured.

Extraction protocol |
Total RNA was isolated using Trizol (Invitrogen) and purified using an RNeasy mini kit (Qiagen, Valencia, CA).

Label protocol |
A total of 5 µg of purified RNA was reverse-transcribed into cDNA using T7-oligo (dT) primer (Affymetrix, Santa Clara, CA) and SuperScript II RT (Invitrogen). The biotin-labeled cRNA was synthesized from cDNA using the BioArray RNA transcript labeling kit (Enzo, Farmingdale, NY) and was followed by fragmentation.

Hybridization protocol |
A total of 15 µg of fragmented cRNA was hybridized on an Affymetrix GeneChip (human U133A) at 45ºC for 16 hours at 60 rpm followed by washing/staining procedures.

Scan protocol |
All probe arrays were scanned using the GeneChip scanner 3000, and operating software (MAS 5.0) was used to scan the images and for data acquisition.
