Title |
FicP adult head, biological rep3 |
Sample type |
RNA |
Source name |
Drosophila adult head
Organism |
Drosophila melanogaster |
Characteristics |
genotype/variation: FicP tissue: decapitulated head genotype: Canton S parents age: Adult
Treatment protocol |
Flies were anaesthetized briefly by using 5% CO2, then immediately dissected. The tissues dissected were the complete head, severed at the neck from the adult flies and the CNS (developing brain), including the optic lobes from the larvae
Growth protocol |
Flies were raised on standard medium on a 12:12 h L:D cycle, at 23C, and at 55% r.h. Flies were collected irrespective of their age and sex, whereas the larvae used were third instar, wandering larvae.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted with RNeasy Mini Kit (Qiagen, Valencia, CA, USA).
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the Genechip Scanner 30007G.
