Extraction protocol |
Total RNA was isolated from each sample using miRNeasy (Qiagen) columns using the protocol provided by Qiagen. Messenger RNA was enriched from about 1 μg of total RNA using the Poly-A enrichment method and cDNA libraries were prepared using an Illumina TruSeq RNA Library kit (Illumina).

Data processing |
The resulting sequences were evaluated for quality using FastQC (version 0.11.9.). Paired end adaptors were trimmed using TrimGalore v.0.6.6. The Ae. aegypti genome sequence (version: AaegL5_2) from Vectorbase was used for mapping and alignments by HISAT2 (version 2.2.1). Raw read indexing and sorting alignments were performed using SAMtools (version 1.11) and count files were generated using HTseq (version 0.11.3).
