Extraction protocol Blood was collected from SLE patients and healthy controls and then processed into serum or plasma which was then screened by array.
Label R-PE conjugated goat anti-human IgG secondary antibody
Label protocol Secondary antibody conjugated to a fluorophore was added to a PBS solution containing conjugated beads bound to antibodies from samples.
 
Hybridization protocol A fluorescent secondary antibody was added to a PBS solution containing conjugated beads bound to antibodies.
Scan protocol A Luminex FlexMAP 3D instrument was used to quantify autoantibody levels in each sample by measuring fluorescence (in MFI) from each barcoded bead.
Description Antibodies in samples bind to barcoded beads conjugated to antigens. Antibody reactivity was quantified by the fluorescence measured as MFI from the secondary antibody.
Data processing Barebeed MFI values were subtracted from the MFI values for all analytes in each sample and replicate MFI values were averaged.
