Mycelia were filtered and zap-frozen in liquid nitrogen prior to RNA extraction.

Growth protocol |
FGSC A4 control samples were grown at 37°C for 22 hours, and oe:AN1599 (PbcR) samples for 26 hours in YES-medium supplemented with 3 % gelatin. pH values of cultures were monitored and samples collected at pH-values ranging from 5.76 to 5.94 indicating the early exponential growth phase.

Extraction protocol |
Total RNA was purified with Qiagen Plant Rneasy Kit from 100 uL of frozen wet mycelia, and DNA was removed by DNase digestion with the RNase-Free DNase set (Qiagen). RNA quality and concentration was determined by analysis with an Agilent 2100 bioanalyzer.

The raw data (.pair file) was subjected to RMA (Robust Multi-Array Analysis; Irizarry et al. Biostatistics 4(2):249), quantile normalization (Bolstad et al. Bioinformatics 19(2):185), and background correction as implemented in the NimbleScan software package, version 2.4.27 (Roche NimbleGen, Inc.).Preliminary data processing was performed by NimbleGen Systems Inc., Madison, WI, USA. Normalized data was further analyzed with ArrayStar software by DNAStar.
