Sample GSM8260660 Query DataSets for GSM8260660
Status Public on May 11, 2024
Title M_control_155pk
Sample type protein

Source name plasma collected after irradiation
Organism Mus musculus
Characteristics strain: C57BL6
tissue: plasma
gender: M
dose: control
timepoint: control
Treatment protocol Murine in vivo models utilized a Precision X-ray source at a dose rate of 407 cGy/min. Dose rate was calibrated based upon the procedures described in American Association of Physicist in Medicine (AAPM) Task Group Report 61 (TG-61) with regard to the following conditions: X-ray tube potential was 320 kV, half value layer (HVL) is 0.8 mm Copper (Cu), source to surface distance (SSD) was 50 cm. Dose rate was measured at 2 cm depth in solid water phantom using a PTW model: N23342 ion chamber and Inovision, model 35040 electrometer.
Growth protocol nine to eleven week old female or male C57BL6 mice received a single total body irradiation (TBI) dose of 2, 4, or 8 Gy or sham radiation. All mice receiving TBI were confined unanesthetized in plexiglass containers. All animal studies were conducted in accordance with the principles and procedures outlined in the NIH Guide for the Care and Use of Animals and procedures were approved by the NIH Lab Animal Safety Program under an approved protocol #ROB170.
Extracted molecule protein
Extraction protocol Approximately 140ul of plasma per sample was used for the Somalogic SomaScan Assay
Label biotin
Label protocol SOMAmerĀ® (Slow Off-rate Modified Aptamer) reagents which consist of short single-stranded DNA sequences that incorporate hydrophobic modifications

Hybridization protocol A SOMAmer-protein binding step is followed by a series of partitioning and wash steps to convert relative epitope concentrations into measurable nucleic acid signals that are quantified using DNA-hybridization microarrays
Scan protocol The cyanine-3 signal from the SOMAmer reagent is detected on microarrays
Description 155pk
Data processing Intraplate and interplate variations were adjusted by SOMAscanĀ® v4 Data Standardization pipeline. Log2(data +1) transformation was performed before all downstream analysis.
