Treatment protocol |
6-9 day old adult female mosquitoes were fed on defibrinated chicken blood experimental glass feeders with 1e7 PFU WNV-Kunjin +/- 170 pM bovine insulin. Mosquitoes were deprived of sucrose 48 hours prior to experimental feedings.

Growth protocol |
Adult female mosquitoes were fed on defibrinated chicken blood in hog sausage casing. Adults were provided continuous access to sucrose.

Animals were homogenized in Trizol reagent. RNA was harvested using Trizol reagent and Direct-Zol RNA extraction kit (Zymo Research). 2.5 ug of total RNA was used for the construction of sequencing libraries. RNA libraries for RNA-seq were prepared using TruSeq Stranded mRNA Library Prep Kit (Illumina) following manufacturer's protocols.

CLC Genomics Workbench v 11.0.1 Sequence reads were imported using Illumina paired importer (FASTQ). Trimmed sequence reads were mapped to GCA_000209185.1 using CLC genomic benchwork (parameters: Discard read names=No, Discard quality scores=No, Paired orientation=Paired reads (forward-reverse), Minimum distance=200, Maximum distance=300, Quality score=NCBI/Sanger or Illumina Pipeline 1.8 and later, Remove failed reads=Yes, MiSeq de-multiplexing=No). Read count extraction and normalization were performed using CLC genomic benchwork.
