Title |
RPE1 cell, control oligo transfect, 72h |
Sample type |
RNA |
Source name |
RPE1 cell transfeted with control oligo for 72h
Organism |
Homo sapiens |
Characteristics |
cell line: hTERT-RPE1 genotype/variation: control
Treatment protocol |
hTERT-RPE1 cells were transfected with control (Ctrl) or miR-129-3p (M129) oligos for 72 h using Lipofectimine RNAiMAX (Invitrogen)
Growth protocol |
hTERT-RPE1 cell lines were cultured with DMEM/F-12 containing 10% FBS and 0.01mg/ml hygromycin B at 37°C, 5% CO2.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted with RNeasy Mini Kit (Qiagen), according to the manufacturer’s instruction.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100ng total RNA (Cat. AM1792, Ambion).
Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45℃ on GeneChip Human Genome U133 Plus 2.0 with constant rotation at the speed of 60 rpm. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
Description |
RPE1-Ctrl.CEL Gene expression data from hTERT-RPE1 cell
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings, and RMA normalization was done using Affy Suit in Bioconductor Package.
