Growth protocol |
HK1 were maintained in RPMI 1640 medium supplemented with 10% fetal calf serum (FCS) while C666-1 in fibronectin coated plate in RPMI 1640 medium supplemented with 15% FCS. For sphere-forming assay, C666-1 cells were seeded at 5000 cells/ well in 6-well low attachment plate in serum-free DMEM/F12 (1:1) media supplemented with 5ug/ml Insulin-Transferrin-Selenium-A, 20ng/ml rEGF, 10ng/ml bFGF, 0.4% BSA, 1xB-27 supplement. All cell lines were maintained in 50U/ml penicillin and 50 µg/ml streptomycin at 37°C in a humidified atmosphere of 5% CO2.

Data processing |
Images from Agilent microarray were processed in Feature Extraction software. Data files from the Agilent chips were imported into GeneSpring as single color analysis and then the signal values transformed to the log base 2. Normalization was carried out with 75th percentile.
