Treatment protocol |
ARPE19 were stimulated basolaterally for 48h with 1) recombinant IFNg (200ng/ml) and/or recombinant TNFa (40ng/ml) or 2) CD3/CD28 activated T cells (2.5 T cells/RPE cell) with anti-IFNg (40ug/ml) and/or anti-TNFa (118ug/ml).

Growth protocol |
The adult human RPE cell line ARPE-19 (American Type Culture Collection) was cultured in 6-well cell culture plates on Anopore 0.2 um membrane inserts (Nunc) at 37 oC with 10% CO2 >6 weeks in DMEM+10%FCS, until pigmented monolayers had formed. Then media was gradually changed to X-vivo 15 serum-free medium (Lonza BioWhittaker) prior to experiments.
