Treatment protocol |
Hypoxia-adapted Drosophila melanogaster were grown in chambers maintained at 4% O2 or reverted to 21% O2 for 5 generations, and generation-matched control flies were maintained in chambers at 21% O2. Cultures were visually inspected and flies were collected from three developmental stages: third instar larvae, 0.5-3 hr post-eclosion adults, and 7-9 day old adults.

Extraction protocol |
Total RNA was extracted from whole flies homogenized in TRIzol (Invitrogen, Carlsbad, CA) followed by a clean-up with RNeasy kit (Qiagen, Valencia, CA).

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 2.5 ug total RNA.

10 ug of fragmented cRNA were hybridized for 16 hr at 45C. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
