Treatment protocol Hypoxia-adapted Drosophila melanogaster were grown in chambers maintained at 4% O2 or reverted to 21% O2 for 5 generations, and generation-matched control flies were maintained in chambers at 21% O2. Cultures were visually inspected and flies were collected from three developmental stages: third instar larvae, 0.5-3 hr post-eclosion adults, and 7-9 day old adults.
Growth protocol Flies were cultured on standard media and maintained in chambers adjusted to 4% or 21% O2, depending on experimental group.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from whole flies homogenized in TRIzol (Invitrogen, Carlsbad, CA) followed by a clean-up with RNeasy kit (Qiagen, Valencia, CA).
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 2.5 ug total RNA.
 
Hybridization protocol 10 ug of fragmented cRNA were hybridized for 16 hr at 45C. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the Affymetrix 7G Scanner.
Description Gene expression data from a pool of 25 male and 25 female 0.5-3 hour post-eclosion adults adapted to 4% O2 and reverted to 21% O2
Data processing Data were analyzed using the Plier algorithm implemented in the Affymetrix Expression Console software using the default settings.
