Treatment protocol |
Infected cells were exposed to viral supernatants of the specified vector in the presence of 6μg/ml polybrene. 48 hours post infection, cells were placed under 1μg/ml puromycin selection.

Cells were grown under standard conditions as specified by the ATCC. 24 hours prior to RNA extraction, cells were seeded at equal confluence.

200ng Total RNA were used. cDNA samples were generated and labeled using the Affymetrix WT-cDNA synthesis and labeling kit.

Hybridization protocol |
2ug ss cDNA in 80 ul were hybridized to a Human Gene 1.0 ST array using Affymetrix hybridization kit materials for 17 hours at 45° at 60rpm. Post-hybridization washes were done with Affymetrix wash solutions on the FS450 station using the FS450_0007 protocol.

Arrays were scanned on the Affymetrix GeneChip Scanner 3000 7G.
