Growth protocol |
Bone marrow fragments from tibia plateau of patients undergoing surgery were flushed with PBS. Cell suspension was filtered through a 100 µm cell strainer and centrifuged for 7 min at 400xg. Cells were seeded into 75 cm2 tissue culture flasks with DMEM-LG (PAA) with 10% FCS (PAA), 2 mM L-glutamine (Sigma-Aldrich) and 100 U/mL penicilline/streptomycine ( Lonza). The first medium exchange was performed after 48 hours to remove erythrocytes and other non-adherent cells. Thereafter, media was exchanged twice per week, and MSC were isolated by their plastic adherence. When reaching 80% confluence, cells were trypsinized, counted in a Neubauer counting chamber (Brand, Wertheim, Germany) and replated at 13,333 cells/cm2.

Data processing |
CEL files were processed using the Affymetrix Power Tools (APT) Software Package. Normalization was done employing RMA (Irizarry et al., Biostatistics 4, 249-264, 2003).
