Treatment protocol |
At ~44 weeks of age, the animals (n=9-10) were gavaged twice daily for 28 days with the specified doses of RAD001 and/or BEZ235. The mice were starved for three hours prior to sacrifice and received their last dose of RAD001 at the time starvation or BEZ235 1 hour before sacrifice.

Growth protocol |
C57Bl/6 mice were obtained from Jackson laboratories. At two weeks of age, male mice were injected intraperitoneally with 50 mg/kg BW of diethylnitrosamine (DEN) (Sigma).

Total RNA from all the samples was purified using the Qiagen RNeasy purification kit following the manufacturer’s protocol.

The integrity of the RNA was measured using a Bioanalyser and RIN values > 7-8 were considered adequate to proceed further with total RNA transcription into cRNA using the Illumina Total Prep RNA amplification kit (Ambion) following the manufacturer’s protocol.

Biotinylated cRNA was hybridized to Illumina MouseRef8v2 bead chips.

Gene expression matrices were scanned using Illumina iSCAN Control Software.
