Title |
Tick-infested mouse, 3 hour after tick attachment, biological rep3 |
Sample type |
RNA |
Source name |
BALB/cJ mouse ear, 4mm biopsy punch
Organism |
Mus musculus |
Characteristics |
strain: BALB/cJ stress: tick infestation time: 3 hr tissue: Skin
Treatment protocol |
Mice were anesthetized and infested with 8 Ixodes scpularis nymphs. Attachment was monitored visually. At the proper times after attachment, mice were euthanized and 4mm biopsy punches from tick attachment sites were collected in RNALater (Ambion).
Growth protocol |
Mice were housed at the University of Texas Medical Branch ABSL-2 animal facility, and fed with food and water ad libitum. All experiments were approved by the UTMB IACUC.
Extracted molecule |
total RNA |
Extraction protocol |
RNA was extracted using a combination of Trizol and Rneasy microkit protocols.
Label |
biotin
Label protocol |
Biotinylated cRNA was prepared using the Ambion MessageAmp™ Premier RNA Amplification Kit from 500nanograms total RNA.
Hybridization protocol |
Total fragmented cRNA (10micrograms) was hybridized to the Affymetrix Mouse GeneChip 430A 2.0 array according to the manufacturer’s (Affymetrix, CA) conditions.
Scan protocol |
Fluorescence was detected with an Affymetrix-7G Gene Array scanner using the Affymetrix GeneChip Command Console software (AGCC1.1).
Description |
Gene expression from mice infested for 3 hrs
Data processing |
Gene expression changes were identified using Partek Genomics Suite (Partek, MO) following the default gene expression workflow.
