Treatment protocol |
Sciatic nerves were dissected, the perineureal sheath was removed, and the nerve were snap-frozen with liquid nitrogen. Prior to RNA extraction, the nerves were pulverized with mortar and pestle, and transferred in the Trizol reagent (Invitrogen).

Sciatic nerves were dissected from WT, CHOP null, S63del amd S63del/CHOP null mice at the indicated timepoints (P5, P28 and 4-months)

Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions. Total RNA was then purified with Rneasy kit (Qiagen) following manufacturer's instruction.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Mouse 430A 2.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G.
