129-derived passenger mutations confound phenotype of ACKR1-deficient mice

(Submitter supplied) A majority of genetically modified mice carry passenger mutations, originating from the 129-derived embryonic stem (ES) cells, near the targeted gene. Unintended retention of these mutations can introduce confounding phenotypes and affect conclusions. We show that Ackr1-/-129ES mice retained approximately 6 Mb of 129 ES cells-derived genomic material in chromosome 1 near Ackr1 in bone marrow derived monocytes (Mono), nucleated erythroblasts (NECs), and polymorphonucleated neutrophils (PMN), notably affecting the expression of PYHIN genes.

Here we demonstrate that glutamine deprivation switch Th17 cells to Treg cells in a time and dose dependent manner. Astrocytic circadian clock control of energy expenditure by transcriptional stress responses in the ventromedial hypothalamus

(Submitter supplied) Mice carrying a mutation which deletes Bmal1 in Gfap-expressing astrocytes, and control animals were fed either a standard diet or a high fat diet for 16 weeks. Conditional deletion of Bmal1 in Gfap cells decreased weight gain and increased energy expenditure under high fat diet, hence the transcriptome of Brown Adipose Tissue (BAT) and Ventromedial Hypothalamus (VMH) were obtained.

Depletion of miR-144/451 alleviates anemia in β-thalassemic mice

(Submitter supplied) Tto investigate whether ablation of miR-144/451 is deleterious for th3+/- mice, we crossed miR-144/451 knockout (mKO) mice (with mild anemia at baseline)13 with th3+/- mice (with severe anemia). We found that mKO/th3+/- double-mutant mice exhibited dramatic improvement in anemia. To explore the mechanism of anemia improvement in mKO/th3+/- mice, we fractionated erythroblasts from bone marrow of four genotypes mice, including WT,mKO, th3+/-,mKO/th3+/- mice,for microarray analysis.
