Replacement of nitrite in meat products by natural bioactive compounds results in reduced exposure to N-nitroso compounds: the PHYTOME project

(Submitter supplied) It has been proposed that endogenously formed N-nitroso compounds (NOCs) are partly responsible for the link between red meat consumption and colorectal cancer (CRC) risk. As nitrite has been indicated as one of the critical factors in the formation of endogenous NOCs, it is of high importance to replace or reduce the nitrite levels in meat. Therefore, the PHYTOME project was initiated (Phytochemicals to reduce nitrite in meat products; www.phytome.eu), an EU funded research project aiming to develop innovative meat products in which the food additive sodium nitrite (E251) has been replaced by natural compounds originating from fruits and vegetables. more...

The gene expression signatures for Huh7 cell lines with HCC-associated HBV variants in small S proteins transfection

(Submitter supplied) To further investigating the novel NGS-defined HCC-associated SNVs in small S region involve hepatocarcinogenesis pathways, the plasmid pEGFP-N1-HBVS-genoB was used as the template for mutagenesis to create mutations T216C, A273G, and double mutant T216C/A273G at small surface (S) region, with the QuikChange site-directed mutagenesis kit (Stratagene, La Jolla, CA). The plasmid pEGFP-N1-HBVS-genoC was used as the template for mutagenesis to create mutations A293G, C446G, A456G, A293G/C446G, C446G/A456G, A293G/A456G, and triple mutant A293G/C446G/A456G at small S region. more...

Gene expression of mesothelioma cell line ACC-Meso-4 treated with shRNA targeting chromobox (CBX) 6, 7, and 8.

(Submitter supplied) Chromobox (CBX) family proteins are components in polycomb repressive complex 1 (PRC1), responsible for targeting PRC1 to the chromatin. We studied genes regulated by CBX6, 7, or 8 in human ACC-Meso-4 mesothelioma cell line by microarray analysis of ACC-Meso-4 cells stably expressing short hairpin RNA (shRNA) targeting CBX-6, 7, 8, or control nontarget shRNA.

Microarray based gene expression profiling of RNA binding protein, RBM10 in HEK 293 cells

(Submitter supplied) To determine the target genes of RBM10,we have employed microarray based gene expression profiling by knocking down RBM10 in HEK 293 cells. Microarray analysis after RBM10 knockdown on HEK 293 cells showed that over 1000 genes were down regulated while another 800 genes up regulated as a result of the knockdown. Among the down regulated genes, we found the significant presence of cardiovascular disease related genes, especially cardiac hypertrophy and heart failure. more...

Expression of genes in THP-1 macrophages in response to C. glabrata wt, Cgvps34Δ and Cgyps(1-11)Δ cells.

(Submitter supplied) To understand the host response of C. glabrata, we have employed the whole genome microarray expression profiling to identify the differentially regulated genes. For this, differentiated THP-1 cells were infected with C. glabrata wt, Cgvps34Δ and Cgyps(1-11)Δ cells for 6 h. Uninfected THP-1 cells, treated with PBS were taken as control. Total RNA was extracted using TriZol Reagent followed by ethonol precipitation. more...

Understanding gene expression signatures with pigmentation changes in Melanocytes

(Submitter supplied) This microarray experiment was performed for identifying the signaling pathways that could be differentially regulated in response to changes in the pigmentation levels in human melanocytes. Melanocytes were either treated with pigmentation inducing agent or depigmenting compound targeting tyrosinase (key enzyme involved in the melanogenesis). The samples were then processed for analyzing the differentially regulated signaling pathway by performing microarray on agilent platform. more...

Profile of gene expression with HCV infection in Huh7.5 cells

(Submitter supplied) Differential expression study of lncRNAs (long non-coding RNAs) and mRNAs in HCV infection. For this we infected Huh7.5 cells with HCV (JFH-1) virus and control cells were mock infected. 48h post infection we isolated total RNA using qiagen RNeasy kit as per manufacturer's protocol. The total RNA was then subjected to microarray for both lncRNAs and mRNAs. The resultant profile can be used to compare gene expression in HCV infection versus mock infected cells.
