The orthologue of the RNA chaperone Hfq is essential for cell motility of the cyanobacterium Synechocystis PCC 6803

(Submitter supplied) The gene ssr3341 was previously suggested to encode an orthologue of the RNA chaperone Hfq in Synechocystis sp. strain PCC 6803. When a phototactic strain of this cyanobacterium was insertionally inactivated at ssr3341, the mutants were not transformable, and were rendered non-phototactic compared to the wild type. The loss of motility was complemented by re-introduction of the wild-type gene, correlated with the re-establishment of type IV pili on the cell surface. more...

The cyanobacterial phytochrome 2 regulates the expression of motility-related genes through the second messenger cyclic di-GMP

(Submitter supplied) The cyanobacterial phytochrome Cph2 is a light-dependent diguanylate cyclase producing the second messenger c-di-GMP. Under blue light, the Cph2-dependent increase in the cellular c-di-GMP concentration leads to inhibition of motility in the cyanobacterium Synechocystis 6803. However, the targets of c-di-GMP in this cyanobacterium and its mechanism of action remained unclear. Here, we determined the cellular concentrations of three cyclic nucleotides in wild-type and Δcph2 cells after blue- and green light illumination. more...

Transcriptomic changes of Synechocystis sp. PCC 6803 upon surface contact after 4 and 8 h

(Submitter supplied) Microarray analysis demonstrated that the expression levels of minor pilin genes change in response to surface contact. A total of 120 genes were determined to have altered transcription between planktonic and surface growth in this analysis. Among these genes, 13 are located on the pSYSM plasmid, and some are newly predicted minor pilin genes.

Gene expression under low O2 in Synechocystis sp. PCC 6803 wild type and hik31 mutant

(Submitter supplied) We have investigated the response of the cyanobacterium Synechocystis sp. PCC 6803 during growth at very low O2 concentration (bubbled with 99.9% N2/0.1% CO2). Significant transcriptional changes upon low O2 incubation included up-regulation of a cluster of genes that contained psbA1 and an operon that includes a gene encoding the two-component regulatory histidine kinase, Hik31. This regulatory cluster is of particular interest, since there are virtually identical copies on both the chromosome and on plasmid pSYSX. more...

Gene expression changes in group 2 sigma factor mutant, ΔsigCDE, in standard growth conditions and after high-light treatment

(Submitter supplied) Adjustment of gene expression during acclimation to stress conditions, such as bright light, in the cyanobacterium Synechocystis sp. PCC 6803 depends on four group 2 σ factors (SigB, SigC, SigD, SigE). A ∆sigCDE strain containing the stress responsive SigB as an only functional group 2 σ factor appears twice as resistant to photoinhibition of photosystem II (PSII) as the control strain. Microarray analyzes of the ∆sigCDE strain indicated that 77 genes in standard conditions and 79 genes in high light were differently expressed compared to the control strain. more...

Genomic responses to copper in Synechocystis

(Submitter supplied) Analysis of genome-wide gene expression patterns in response to copper in WT and COPR mutant in the cyanobacterium Synechocystis sp PCC 6803. Here we have used microarrays to interrogate the global responses to copper additions at non-toxic (0.3 micromolar) and toxic concentrations (3 micromolar) of copper in WT and in a mutant in the copR gene. Addition of the non-toxic copper concentration stimulated the metabolism and induced the switch in the use from cyitochrome c6 to plastocyanin. more...

Gene expression patterns of sulfur starvation in Synechocystis sp. PCC 6803

(Submitter supplied) The unicellular cyanobacterium Synechocystis sp. PCC 6803 is a model system for studying biochemistry, genetics and molecular biology of photobiological processes. Despite its importance in basic and applied research, the genome-wide picture of transcriptional regulation in this bacterium is limited. Characteristic transcriptional responses to changes in the growth environment are expected to provide a scaffold for describing the Synechocystis transcriptional regulatory network as well as efficient means for functional annotation of genes in the genome. more...

Deletion of the Synechocystis sp. PCC 6803 kaiAB1C1 gene cluster causes impaired cell growth under light-dark conditions

(Submitter supplied) In contrast to Synechococcus elongatus PCC 7942, which has been the model cyanobacterium for the study of the prokaryotic circadian clock for more than 20 years, only few data exist on the circadian behaviour of the widely used cyanobacterium Synechocystis sp. PCC 6803. The standard kaiABC operon present in this organism was shown to encode a functional KaiC protein which interacts with KaiA, similar to the Synechococcus elongatus PCC 7942 clock. more...

Integrated transcriptomic and metabolomic characterization of the low-carbon response using an ndhR mutant of Synechocystis sp. PCC 6803

(Submitter supplied) The acquisition and assimilation of inorganic carbon (Ci) represents the largest flux of inorganic matter in photosynthetic organisms, and, hence it is tightly controlled. We studied the Ci-dependent transcriptional and metabolic regulation in the Synechocystis sp. PCC 6803 wild type compared to a DndhR mutant that is defective in the main transcriptional repressor for Ci acquisition genes, NdhR (CcmR). more...

CyAbrB2 contributes to the transcriptional regulation of low CO2 acclimation in Synechocystis sp. PCC 6803

(Submitter supplied) Acclimation to low CO2 conditions in cyanobacteria involves the coordinated regulation of genes mainly encoding components of the carbon concentration mechanism (CCM). Making use of several independent microarray datasets a core set of CO2-regulated genes was defined for the model strain Synechocystis sp. PCC 6803. On the transcriptional level, the CCM is mainly regulated by the well-characterized transcriptional regulators NdhR and CmpR, whereas the role of an additional regulatory protein, namely cyAbrB2 belonging to the widely distributed AbrB regulator family that was originally characterized in the genus Bacillus, is less defined. more...

Slr0643, a S2P Homolog, Is Essential for Acid Acclimation in Cyanobacterium Synechocystis sp. PCC 6803

(Submitter supplied) Every cyanobacterial species contains gene encoding site-2-protease (S2P) homolog. The studied prokaryotic S2P homologs play essential roles in regulating stress response through intramembrane proteolysis of membrane-bound anti-sigma factors. Here, gene of Slr0643, one of four S2P homologs in Synechocystis sp. PCC 6803, was insertionally disrupted to explore its physiological role. Only a partially segregated mutant was obtained, indicating its indispensability for growth. more...

Evidence for a major role of antisense RNAs in cyanobacterial gene regulation

(Submitter supplied) The model cyanobacterium Synechocystis sp. PCC 6803 was used for a systematic survey on the number and types of antisense (as)RNAs. A tiled microarray was constructed with probes for both strands of the genes or intergenic spacers with candidate transcripts predicted by an algorithm and an equally sized and distributed control set. The resulting 102,739 individual probes cover an accumulated length of 1,441,146 nt or about 40% of the total chromosome sequence of Synechocystis 6803. more...
