Lin28b promotes fetal B lymphopoiesis through the transcription factor Arid3a [wildtype tissue]

(Submitter supplied) Mouse B cell precursors from fetal liver and adult bone marrow generate distinctive B cell progeny when transplanted into immunodeficient recipients, supporting a two-pathway model for B lymphopoiesis, fetal “B-1” and adult “B-2”. Recently Lin28b was shown to be important for the switch between fetal and adult pathways; however, neither the mechanism of Lin28b action nor the importance of BCR signaling in this process was addressed. more...

The Genetic Networks that Orchestrate Development of Specific Waves of Fetal and Adult B-1 and B-2 Development are Distinct

(Submitter supplied) B lymphocyte development is often depicted as a linear process that initiates in the fetus and continues in the bone marrow after birth. Based on the dissection of B lymphopoiesis in a PU.1 hypomorphic strain of mice, generated by deletion of a 14kb upstream regulatory element of the Sfpi1 gene, and the transcriptional profiling of fetal and adult B cell progenitors, we establish the existence of three waves of B-1 and two waves of B-2 development and show that each of these waves has distinct transcriptional requirements. more...

Assessment of ability of hTSLP to maintain primary CRLF2 B-ALL cells in a xenograft model in a state more similar to the parent leukemia

(Submitter supplied) Xenograft models represent an excellent method for expanding primary leukemias, but their ability to preserve the gene expression profile of the parent leukemia may also depend on providing microenvironmental factors that are not cross-reactive between human and mouse. Here we focused on leukemias with a CRLF2 mutation, and the ability of human TSLP to stimulate these cells.

TSLP-Induced Gene Expression Signature in Primary CRLF2 B-ALL

(Submitter supplied) The cytokine TSLP stimulates in vitro proliferation of human fetal B cell progenitors. Genetic alterations that cause overexpression of the TSLP receptor component, CRLF2, lead to B cell acute lymphoblastic leukemia (CRLF2 B-ALL) with poor outcome. The in vivo role of TSLP in normal human B cell development and its contribution to CRLF2 B-ALL are unknown. In classic xenograft models CRLF2-mediated signaling is unlikely to be activated by mouse TSLP, based on data from engineered cellular models. more...

Stage-specific control of early B cell development by the transcription factor Ikaros

(Submitter supplied) Ikaros is an essential regulator of lymphopoiesis. Here, we studied the B-cell-specific function of Ikaros by conditional Ikzf1 inactivation in pro-B cells. B-cell development was arrested at an aberrant ‘pro-B’ cell stage characterized by increased cell adhesion and loss of pre-B cell receptor signaling. Ikaros was found to activate genes coding for pre-BCR signal transducers and to repress genes involved in the downregulation of pre-BCR signaling and upregulation of the integrin signaling pathway. more...

Lin28b augments protein synthesis to control B cell positive selection early in life [RIP-seq]

(Submitter supplied) In this study, we identify elevated protein synthesis as a defining characteristic of early life B cell development and a determinant for the qualitative switch in B cell output whereby self-reactive CD5+ B-1 cells becomes censored in postnatal mice This is in part mediated by the heterochronic RNA binding protein LIN28B. To identify Lin28b-dependent, stage specific effects, B cell progenitors at different developmental stages were sorted and analyzed by RNA-seq

Single-cell RNA expression profiling of mouse common lymphoid progenitors with or without dox-induction of LIN28B
