Exposure of bovine oocytes and embryos to elevated non-esterified fatty acid concentrations: integration of epigenetic and transcriptomic signatures in resultant blastocysts [gene expression]

(Submitter supplied) Metabolic stress associated with negative energy balance in high producing dairy cattle and obesity in women is a risk factor for decreased fertility. Non-esterified fatty acids (NEFA) are involved in this pathogenesis as they jeopardize oocyte and embryo development. Growing evidence indicates that maternal metabolic disorders can disturb epigenetic programming, and more specifically DNA methylation, in the offspring. more...

Good quality day 7 embryos display disturbed cellular functions during post-hatching development when oocytes were matured under lipotoxic conditions: novel insights from a bovine embryo-transfer model

(Submitter supplied) Objective: To study post-hatching development after transfer of morphologically good quality embryos that survived in vitro maturation under lipolytic conditions using a well-established and recognized bovine model. Design: prospective study with a crossover design. Setting: research laboratory. Animals: 8 healthy multiparous non-lactating Holstein cows used in repeated embryo transfers. Intervention(s): Bovine cumulus oocyte complexes (COCs) were in vitro matured in the presence of either 1) a combination of elevated pathophysiological concentration of palmitic acid (PA) and physiological concentrations of stearic acid (SA) and oleic acid (OA) (HIGH PA), or 2) physiological concentrations of PA, SA and OA (BASAL). more...

Gene expression analysis of in vivo originated bovine embryos fllushed out at 2- or 8-cell stages and cultured in vitro till blastocyst stage

(Submitter supplied) There is an inevitable need for combining in vitro and in vivo culture systems during specific developmental time points to facilitate a comprehensive understanding of early embryo development which would yield insights into the molecular pathways controlling early development and to improve our knowledge in regulation of embryonic development. In current study, we aimed to understand the influences of in vitro culture conditions on bovine blastocyst originated in vivo at 2- or 8-cell stagre. more...

Comparative analysis of sperm DNA methylation supports evolutionary acquired epigenetic plasticity controlling organ speciation

(Submitter supplied) Gametic DNA methylation is essential to the viability of the embryo and participates in developmental programming. Here, we performed a comparative epigenomic analysis of DNA methylation in spermatozoa from humans, mice, rats and mini-pigs to get insight into species specificity of the gametic epigenetic blueprint. Using data from whole-genome bisulfite sequencing (WGBS) and reduced representation bisulfite sequencing (RRBS), we compared the methylation profiles of orthologous single-CpG sites. more...

Metabolic Stress Induces Modifications in the Epigenetic Program of Preimplantation Bovine Embryos

(Submitter supplied) we induced an energetic stress on in vitro embryos to observe the epigenetic response associated with metabolic stress. Embryos were created with slaughterhouse cows oocytes and then exposed to an elevated glucose concentration (5mM vs 0.2mM for the control embryos) for the first 3 days post fecundation and then cultivated in normal media until blastocyst stage. Blastocysts were collected from 4 different weeks of culture. more...

Single-cell DNA methylation sequencing reveals epigenetic variations in mouse oocyte superovulated with different doses of FSH/hMG

(Submitter supplied) Background: With the rapid development of assisted reproductive technology (ART), although ovulation induction has made great progress, the accompanying health risks should not be ignored. Epigenetic modifications play an important role during the development of gametes and embryos. Recently, the epigenetic abnormalities caused by superovulation have attracted increasing attention. The follicle-stimulating hormone (FSH) and human menopausal gonadotropin (hMG) are common gonadotropins used for superovulation and appropriate concentration of them might be necessary. more...

Dynamics of mitochondrial DNA methylation in oocytes, embryos and granulosa cells

(Submitter supplied) Comparison of mitochondrial DNA (mtDNA) methylation patterns in oocytes, blastocysts and ovarian granulosa cells indicates hitherto unsuspected dynamics. Oocytes and blastocysts recovered from animals subjected to ovarian stimulation and from abattoir ovaries were analyzed using bisulphite transformation of DNA followed by whole genome sequencing. The number of mtDNA copies is quite high in oocytes (200–400,000) and early embryos, resulting in very high coverage (> 3000 X) and very low p values for each of 716 cytosine-based nucleosides. more...
