In planta bacterial RNA-Seq with bacterial isolation.

(Submitter supplied) Plant pathogens can cause serious diseases that impact global agriculture1. Understanding how the plant immune system naturally restricts pathogen infection holds a key to sustainable disease control in modern agricultural practices. However, despite extensive studies into the molecular and genetic basis of plant defense against pathogens since the 1950s2,3, one of the most fundamental questions in plant pathology remains unanswered: how resistant plants halt pathogen growth during immune activation. more...

Multidimensional gene regulatory landscape of a bacterial pathogen in plants

(Submitter supplied) Understanding the gene regulation of plant pathogens is crucial for pest control and thus global food security. An integrated understanding of bacterial gene regulation in the host is dependent on multi-omic datasets, but these are largely lacking. Here, we simultaneously characterized the transcriptome and proteome of a bacterial pathogen in plants. We found a number of bacterial processes affected by plant immunity at the transcriptome and proteome levels. more...

Global regulatory networks active in Pseudomonas syringae B728 during leaf colonization and in response to target environmental stresses: the GacS, SalA, RetS, AlgU, HrpL, RpoN, RpoS, AhlR and AefR regulons

(Submitter supplied) The plant pathogen Pseudomonas syringae pv. syringae B728a grows and survives on leaf surfaces and in the leaf apoplast of its host, bean. Global transcriptome profiling was used to understand the contribution of distinct regulators to B728a fitness and pathogenicity. We performed a transcriptome analysis of B728a and nine regulator mutants recovered from the surface and interior of leaves and exposed to various environmental stresses in culture. more...

Modeling regulatory relationships in Arabidopsis immune signaling network.

(Submitter supplied) Expression profiles of 22 reference Arabidopsis immunity mutants were collected using the Arabidopsis Pathoarray 464_001 (GPL3638) in order to build a network model predicting the Arabidopsis immune signaling network. Biological signaling processes may be mediated by complex networks in which network components and network sectors interact with each other in complex ways. Studies of complex networks benefit from approaches in which the roles of individual components are considered in the context of the network. more...

Gene expression profiling study by RNA-seq in Arabidopsis thaliana infected by Vibrio vulnificus

(Submitter supplied) The objective of this study is to assess gene expression changes in Arabidopsis thaliana as time passed after Vibrio vulnificus 96-11-17M infection. We performed transcript profiling using 10 RNA-seq samples of Arabidopsis obtained at 0h, 12h, 24h, 48h, and 72h after 96-11-17M treatment. Through various statistical tests, differentially expressed genes at each time point were obtained and enriched biological functions were compared across infection time points. more...

The infiltrated cells were harvested in parallel with the immediately adjacent uninfected cells. Samples were collected at 0,1,2,4,6 post-inoculation in three biological replicates.

Escalation in the host-pathogen arms race: a host resistance response corresponds to a heightened bacterial virulence response

(Submitter supplied) The zig-zag model of host-pathogen interaction describes the relative strength of defense response across a spectrum of pathogen-induced plant phenotypes. A stronger defense response results in increased resistance. Here, we investigate the strength of pathogen virulence during disease and place these findings in the context of the zig-zag model. Global transcriptional analysis reveals unique and shared responses in Arabidopsis thaliana exposed to combined drought and pathogen stress

(Submitter supplied) With frequent fluctuations in global climate, plants often experience co-occurring dry-wet cycles and pathogen infection and this combination adversely affects plant survival. In the past, some studies indicated that morpho-physiological responses of plants to the combined stress are different from the individual stressed plants. However, interaction of drought stressed or drought recovered plants with pathogen has not been widely studied at molecular level. more...

Interactions between effector-triggered immunity (ETI) and pattern-triggered immunity (PTI) in an Arabidopsis dde2 ein2 pad4 sid2 mutant

(Submitter supplied) We observed inhibition of the hypersensitive response (a typical ETI response) by PTI signaling in an Arabidopsis quadruple mutant dde2 ein2 pad4 sid2 (quad). Thus, we designed an experiment to see the interaction between ETI and PTI signaling at the transcriptome level. The Arabidopsis line dde2 ein2 pad4 sid2 Ed-AvrRpt2 (quadAvrRpt2) was used (Ed-AvrRpt2, estradiol-inducible AvrRpt2 transgene). In this plant line, ETI can be elicited by estradiol (Ed) treatment via in planta expression of AvrRpt2. more...

Cell type identity determines transcriptomic immune responses in Arabidopsis thaliana roots

(Submitter supplied) Root pathogens are a major thread in global crop production and protection strategies are required to sustainably enhance the efficiency of root immunity. Our understanding of root immunity is still limited in comparison to the knowledge gained for the regulation of immune response in leaves. In an effort to reveal the organisation of root immunity in roots, we undertook a cell type-specific transcriptome analysis to identify gene networks in epidermis, cortex and pericycle cells of Arabidopsis roots upon treatment with two immunity elicitors, bacterial microbe-associated molecular pattern flagellin and the endogenous damage-associated molecular pattern Pep1. more...

Pathogen induced m6A dynamics regulate plant immunity

(Submitter supplied) Post-transcriptional regulation of mRNA mediated by methylation at the N6 position of adenine (N6-methyladenosine (m6A)) can have profound effects on transcriptome regulation in plants. Focused studies across eukaryotes offer glimpses into the processes governed by m6A throughout developmental and disease states. However, we lack an understanding of the dynamics and scope of regulatory potential m6A possesses during biotic stress responses in plants. more...
