MTG16 regulates colonic epithelial differentiation, colitis, and tumorigenesis by repressing E protein transcription factors

(Submitter supplied) Aberrant epithelial differentiation and regeneration contribute to colon pathologies, including inflammatory bowel disease (IBD) and colitis-associated cancer (CAC). Myeloid translocation gene 16 (MTG16, also known as CBFA2T3) is a transcriptional corepressor expressed in the colonic epithelium. MTG16 deficiency in mice exacerbates colitis and increases tumor burden in CAC, though the underlying mechanisms remain unclear. more...

Kaiso is required for MTG16-dependent effects on colitis-associated carcinoma

(Submitter supplied) The myeloid translocation gene family member MTG16 is a transcriptional corepressor that relies on the DNA-binding ability of other proteins to determine specificity. One such protein is the ZBTB family member Kaiso, and the MTG16:Kaiso interaction is necessary for repression of Kaiso target genes such as matrix metalloproteinase-7. Using the azoxymethane and dextran sodium sulfate (AOM/DSS) murine model of colitis-associated carcinoma, we previously determined that MTG16 loss accelerates tumorigenesis and inflammation. more...

Expression data from intestinal epithelial cells (IECs) [Mouse430_2 array]

(Submitter supplied) Polycomb group (PcG) proteins are epigenetic silencers whose dysregulation is frequently linked to cancer via mechanisms that remain unclear. Using conditional knock-out mice in a colitis-associated colorectal cancer (CAC) model, we found that Bmi1 and Mel18 are important initiation and maintenance factors during CAC tumorigenesis. Epithelial depletion of both Bmi1 and Mel18, but not either gene alone, significantly reduces tumor growth and multiplicity. more...

Expression data from distal colon tissues of WT and Gm31629-/- mice

(Submitter supplied) Mucosal healing is a key treatment goal for inflammatory bowel disease, and epithelial regeneration is required for an intact gut epithelium. Long noncoding RNAs are involved in the development of inflammatory bowel disease. Here, we investigated the role of a novel long noncoding RNA, Gm31629, in epithelial regeneration. We used RNA-seq to to detail the global programme of gene expression in distal colon tissues from WT and Gm31629-/-.

FOXO3 Deficiency in Neutrophils Drives Colonic Inflammation and Tumorigenesis

(Submitter supplied) Inflammatory bowel disease (IBD), characterized by infiltration of polymorphonuclear neutrophils (PMNs), increases the risk of colon cancer. PMN activation corresponds to accumulation of their intracellular Lipid Droplets (LDs). As increased LDs are negatively regulated by transcription factor FOXO3, our aim is to determine the significance of this regulatory network in PMN-mediated IBD and tumorigenesis. more...

A temporal classifier predicts histopathology state and parses acute-chronic phasing in inflammatory bowel disease patients

(Submitter supplied) We characterized the transcriptional landscape of acute and chronic models of dextran sodium sulfate (DSS) and adoptive transfer (AT) colitis murine models to derive novel temporal gene expression and splicing signatures in blood and colonic tissue in order to capture dynamics of colitis remission and relapse. We show that colitis induces robust expression of minor isoforms and using machine learning, demonstrate that specific temporal gene expression and splicing signatures are highly predictive of tissue histopathology spanning blood and colonic tissue as well as various experimental sites and protocols. more...
