Response to low oxygen conditions

Analysis of Pseudomonas aeruginosa strain PAO1 grown with 2% or 0.4% oxygen (O2), or without O2 but with nitrate instead. In cystic fibrosis (CF) lungs, PAO1 grows to high densities in mucopurulent material depleted in O2. Results provide insight into mode of growth in CF lungs.

Responses of Pseudomonas aeruginosa to low oxygen

(Submitter supplied) In order to understand how Pseudomonas aeruginosa responds to low oxygen we grew strain PAO1 with 3 different oxygen concentrations: 2%, 0.4% and 0% supplemented with nitrate as an electron acceptor. Gene expression under these conditions was compared to that of cells grown with 20% oxygen. Keywords: Comparison of transcriptome profiles

Gene expression in AES-2 clonal isolates of Pseudomonas aeruginosa

(Submitter supplied) Pseudomonas aeruginosa (P. aeruginosa) lung infection is a significant cause of mortality in patients with cystic fibrosis (CF). Most CF patients acquire unique P. aeruginosa strains from the environment; however clonal strains have been identified in CF communities in several countries. Two clonal strains infect 10% to 40% of patients in three CF clinics in mainland eastern Australia. The expression profiles of four planktonically-grown isolates of one Australian clonal strain (AES-2), and four non–clonal CF P. more...

Expression data from sequential P. aeruginosa cystic fibrosis (CF) isolates

(Submitter supplied) To provide a more detailed survey of adaptive changes in the physiology of P. aeruginosa (PA) during long-term infection of the cystic fibrosis (CF) lung, we performed a comparative proteome and transcriptome analysis of a set of isogenic sequential non-mutator and mutator isolates from three selected CF patients. Recently, we showed that during CF lung persistence PA mutators converge to a virulence-attenuated phenotype. more...

Transcriptome analysis of the roxSR and anr mutant strains of Pseudomonas aeruginosa under aerobic conditions

(Submitter supplied) To assess the role of two redox-sensitive transcriptional regulators, RoxSR and ANR, in Pseudomonas aeruginosa under aerobic conditions, microarray analysis was performed. Transcriptome profiles of roxSR mutant and anr mutant aerobically grown in LB medium were determined by Affymetrix GeneChip at both the exponential phase and early stationary phase and compared to that of the wild type strain.

Using RNA Seq to define the regulon of the Pseudomonas aeruginosa transcription factor Anr in low-oxygen colony biofilms.

(Submitter supplied) Purpose : The goal of this study was to use RNA Seq to define the regulon of the transciption factor Anr by comparing global transcriptional profiles of Pseudomonas aeruginosa strain PAO1 and a clinical isolate with their isogenic ∆anr mutants, grown in colony biofilms at 1% oxygen. Methods : mRNA profiles were generated for laboratory strain PAO1 and for a clinical isolate J215, as well as for ∆anr derivatives of each strain, in duplicate, by deep sequencing. more...

The anti-sigma factor MucA of Pseudomonas aeruginosa: dramatic differences of mucA22 vs. DmucA mutants in anaerobic acidified nitrite sensitivity of planktonic and biofilm bacteria in vitro and during chronic murine lung infection

(Submitter supplied) In this study, the A-NO2- sensitivity of mucA22 mutant bacteria vs. a genome sequence-confirmed mucA deletion (∆mucA) mutant was determined. The ∆mucA is surprisingly more resistant to A-NO2- than mucA22 bacteria. The genes responsible for the disposal of nitric oxide (NO), a by-product of A-NO2-, were found to be near wild-type levels in the ∆mucA mutant yet were dramatically reduced in the mucA22 mutant.

Effects of vitamin B12 on the gene expression of PAO1 under anaerobic growth conditions

(Submitter supplied) Pseudomonas aeruginosa undergoes cell elongation and forms robust biofilms during anaerobic respiratory growth using nitrate (NO3-) as an alternative electron acceptor. Understanding the mechanism of cell shape change induced upon anaerobiosis is crucial to the development of effective treatments against P. aeruginosa biofilm infection. Anaerobic growth of PAO1 reached higher cell density in the presence of vitamin B12, an essential coenzyme of class II ribonucleotide reductase. more...

Using RNA Seq to identify a CF habitat specific transcriptional profile

(Submitter supplied) Purpose: The goal of this study was to use RNA Seq to explore whether and to what extent genetic heterogeneity would shape the transcriptional profile in the environment of the CF lung Methods : mRNA profiles were generated for Pseudomonas aerugionsa samples derived from explanted lung tissue or pure cultures isolated from the same lung regions by deep sequencing. To enrich the bacterial RNA MicrobeEnrich Kit (Ambion) was used. more...

P. aeruginosa PA14 gene expression in artificial sputum medium (ASMDM)

(Submitter supplied) Pseudomonas aeruginosa airway infection is the leading cause of morbidity and mortality in cystic fibrosis (CF) patients. In vitro models that closely mimic CF sputum are needed to improve understanding of the pathobiology of P. aeruginosa in the CF airway. We developed an artificial sputum medium (ASMDM) that more closely resembles the composition of CF sputum than current media. In order to validate the utility of ASMDM, we used GeneChip microarrays to compare expression data of P. more...

(Submitter supplied) To investigate the gene expression profile of pellicle cells of Pseudomonas aeruginosa, microarray analysis was performed. Transcriptome profiles of pellicle cells and planktonic cells grown in LB medium were determined by Affymetrix GeneChip.
