Furin deficiency effect on T-cells

Analysis of T-cells lacking the proprotein convertase furin. Proprotein convertases promote the proteolytic maturation of proproteins. Furin is induced in activated T-cells. Results provide insight into the function of furin in T-cells.
Organism:
Mus musculus
Type:
Expression profiling by array, count, 2 genotype/variation sets

T cell-expressed proprotein convertase furin is essential for maintenance of peripheral tolerance

(Submitter supplied) Furin is a proprotein convertase induced in activated T cells, reported to processes the anti-inflammatory cytokine TGFb-1. Herein, we show that conditional deletion of furin in T cells allowed for normal T cell development but impaired the function of regulatory T cells and effector cells, which produced less TGFb-1. Furin-deficient Treg cells, were less protective in a T cell transfer colitis model and failed to induce Foxp3 in normal T cells. more...

Comparison of wild type “empty” and Furin super-enhancer deleted SE EL-4 cell lines

(Submitter supplied) Exceedingly abundant p300 histone acetylase binding was reported upstream Furin in mouse Th1 cells. We aimed to investigate the role of this genomic area in RNA expression by deleting it from EL-4 mouse thymoma cells and comparing the RNA expression profiles in steady-state and after PMA/ionomycin stimulation.

CD69-dependent gene expression in activated CD4 T cells from the spleen of Mus musculus

(Submitter supplied) CD69 is a transmembrane protein expressed on the surface of activated leukocyte. The ligand for CD69 and the intracellular signaling pathway of this molecule are yet unknown. It is widely used as a marker of activated lymphocyte, but its function in immune system is not known. We used micro-array to define genes whose expression is regulated by activation antigene CD69.

CD8+ T cell mediated lung inflammation

(Submitter supplied) Role for naturally occurring CD4+CD25+Foxp3+ regulatory T cells (nTregs) in counterbalancing this process. Using a transgenic murine model for autoimmune-mediated lung disease, we demonstrated that, despite pulmonary inflammation, lung-specific CD8+ T cells can reside quiescently in close proximity to self-antigen. Whereas self-reactive CD8+ T cells in the inflamed lung and lung-draining lymph nodes down-regulated the expression of effector molecules, those located in the spleen appeared to be partly antigen-experienced and displayed a memory-like phenotype. more...

IL-2Rβ- and CD103-dependent genes in regulatory T cells in the gut mucosa

(Submitter supplied) This study determined the genes that are differentially expressed when regulatory T cells (Tregs) were isolated from the lamina propria of the small and large intestine from mice with impaired IL-2Rβ signaling (designated Y3) or impaired IL-2Rβ signaling and lack of CD103 expression (designated Y3/CD103-/-) when compared to Tregs from WT mice. 204 unique annotated mRNAs were differentially expressed by ≥1.5 fold between these 3 groups (Fig. more...

The chromatin-modifying enzyme Ezh2 is critical for the maintenance of regulatory T cell identity after activation

(Submitter supplied) Regulatory T cells (Tregs) are essential for maintaining proper immune homeostasis. Extracellular signals (e.g. TCR, CD28, IL-2R) are necessary for the generation and maintenance of Tregs, but how these signals are integrated to control the gene expression patterns of Tregs is less clear. Here we show that the epigenetic regulator, Ezh2, was induced by CD28 costimulation and Ezh2 activity was elevated in Tregs as compared to conventional CD4+ T cells. more...

Single-cell RNA-seq analysis of WT and Furin KO TECs

(Submitter supplied) To assess the effect of Furin ablation on TEC development and heterogeneity, we detected the transcriptional profiles of 2-week-old WT and Furin KO TECs using single-cell RNA-seq. Employing an unsupervised graph-based clustering strategy, 20 clusters of TECs and 5 clusters of non-TEC contaminants were identified. After removing contaminants, 4301 WT TECs and 4086 Furin KO TECs were obtained. UMAP visualization revealed that the proportion of mTECs decreased significantly and the proportion of cTECs increased significantly in TEC-specific Furin knockout mice in comparison with WT mice. more...
