Transcriptional regulation of plant biomass degradation and carbohydrate utilization genes in Caldicellulosiruptor bescii [RNA-Seq (TSS)]

(Submitter supplied) Extremely thermophilic bacteria from the genus Caldicellulosiruptor can degrade various polysaccharide components of plant cell walls. Previous experimental studies identified a variety of carbohydrate-active enzymes in model species C. saccharolyticus and C.bescii, while prior transcriptomic experiments identified their putative carbohydrate uptake transporters. We investigated the mechanisms of transcriptional regulation of carbohydrate utilization genes using a comparative genomics approach applied to fourteen Caldicellulosiruptor species. more...

Membrane potential limits growth and substrate utilization in C. bescii, alleviation dramatically improves productivity

(Submitter supplied) Caldicellulosiruptor bescii is an anaerobic hyper thermophile that can utilize a wide range of substrates. However, inhibitors released from biomass can result in unfavorable growth conditions and limit bioconversion to products. Medium as well as intracellular pH are conditions critical for growth and prone to change in effect of fermentation end or by products such as, CO2, organic acids etc. Growth pH for C. more...

Metabolic and Expression Regulation in Caldicellulosiruptor bescii

(Submitter supplied) Two single-gene knockout strains (ΔfapR and Δcra) and their common parent strain (JWCB005) were each grown in three conditions of elevated osmolarity which produced a moderate growth defect in all strains and relative growth phenotypes between the strains.

Comparative analysis of extremely thermophilic Caldicellulosiruptor species reveals common and differentiating cellular strategies for plant biomass utilization

(Submitter supplied) Microbiological, genomic and transcriptomic analyses were used to examine three species from the bacterial genus Caldicellulosiruptor with respect to their capacity to convert the carbohydrate content of lignocellulosic biomass at 70°C to simple sugars, acetate, lactate, CO2 and H2. C. bescii, C. kronotskyensis and C. saccharolyticus solubilized 38%, 36% and 29% (by weight) of unpretreated switchgrass (5 g/l), repectively, which was about half of the concentration of crystalline cellulose (Avicel, 5 g/l) that was solubilized under the same conditions. more...

Phylogenetic, microbiological and glycoside hydrolase diversities within the extremely thermophilic, plant biomass-degrading genus Caldicellulosiruptor

(Submitter supplied) Phylogenetic, microbiological and comparative genomic analysis was used to examine the diversity among members of the genus Caldicellulosiruptor with an eye towards the capacity of these extremely thermophilic bacteria for degrading the complex carbohydrate content of plant biomass. Seven species from this genus (C. saccharolyticus, C. bescii (formerly Anaerocellum thermophilum), C. hydrothermalis, C. more...

NCSU C. bescii 3K v1.0

(Submitter supplied) A DNA microarray was designed and constructed based on 2674 protein-coding sequences in the C. bescii genome obtained from the Department of Energy's Joint Genomics Institute (https://img.jgi.doe.gov). OligoArray 2.1 (Rouillard, 2003) was used to generate one 30 to 60-mer oligonucleotide probe sequence for each open reading frame. The probes were synthesized (Integrated DNA Technologies, IA), re-suspended in 50% DMSO, and printed onto Ultragap microarray slides (Corning, NY) using a QArrayMini arrayer (Genetix, UK). more...
