Sample GSM116100 Query DataSets for GSM116100
Status Public on Jun 24, 2006
Title hTERT_EXTENDED_HD2_REP3
Sample type RNA

Source name CD8 T lymphocytes
Organism Homo sapiens
Characteristics late passage CD8 T cells expressing hTERT
Treatment protocol Mycoplasma-free cells were harvested after 15 to 18 days of mitogen stimulation (PHA), Annexin-V expression <10%.
Growth protocol Cell are cultures in RPMI 1640 medium supplemented with 8% Human Serum and 150U/ml of rIL-2 (GlaxoSmithKline) and stimulated using PHA (Sodiag) and irradiated feeder cells.
Extracted molecule total RNA
Extraction protocol RNA from 30x10E6 cells was extracted, purified and used for cDNA synthesis following standard procedures (www.Affymetrix.com).
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 micrograms total RNA.

Hybridization protocol Following fragmentation, 10 micrograms of cRNA were hybridized for 16 hr on human U133Plus 2.0 GeneChip, Chips were washed and stained in the Fluidics Station 400.
Scan protocol GeneChips were scanned using the Hewlett-Packard GeneArray Scanner.
Description hTERT expressing transduced CD8 T Lymphocytes, early passage, healthy donor 2, rep3
Data processing The .cel files data were analyzed with the R library affy, using RMA default settings and the quantile normalization method.
