Treatment protocol Ten 10-11 weeks old C57BL6 mice were ovariectomized under isoflurane-induced anesthesia and sacrified seven days after GDX. 24h before sacrifice, mice were injected s.c vehicle solution (Control group).
Growth protocol Ten to eleven week-old female C57BL6 mice were received from Charles River (St Constant, Canada) and were allowed to acclimate for 4 weeks. The animals were housed individually in an environmentally-controlled room (temperature: 22 ± 3°C; humidity: 50 ± 20%; 12-h light-12-h dark cycles, lights on at 07:15h). The mice had free access to tap water and a certified rodent feed (Lab Diet 5002 (pellet), Ralston Purina, St Louis, MO).
Extracted molecule total RNA
Extraction protocol Mice were anesthetized by inhalation of isoflurane and exsanguinated via cardiac puncture followed by cervical dislocation. Then, total uterus was collected and any adhering tissue was removed. Tissue was then rapidly frozen in liquid nitrogen and kept at -80C until RNA extraction. Trizol extraction of total RNA was performed from pooled tissues, according to the manufacturer's instructions (Invitrogen).
Label Biotin
Label protocol Biotinylated cRNA was prepared according to the standard Affymetrix protocol from 20 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
 
Hybridization protocol Following fragmentation, 15 ug of cRNA was hybridized for 16 hr at 45C on GeneChip MOE 430 2.0 Mouse Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the Affymetrix Scanner 3000 G7.
Description Analysis of uteri of 10-11 weeks old C57Bl/6 females sacrified 24h after a single injection of vehicle (Control animal).
Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 500.
