Title |
Longissimus cervicisMuscle_<1hrPostMortem_rep2 (7731) |
Sample type |
RNA |
Source name |
Muscle tissue collected < 1hr post-mortem before electrical-stimulation
Organism |
Bos indicus x Bos taurus |
Characteristics |
residual shear force (es): LOW tissue: Longissimus cervicis gender: Steer age: aprx. 18month
Extracted molecule |
total RNA |
Extraction protocol |
1. RNA was extracted from aprx. 100-200 mg muscle sample using TRI Reagent and BCP (Molecular Research Center) 2. RNA was tested for quality using Agilent 2100 Series Bioanalyzer 3. RNA with RIN > 8.0 were treated with DNase (Invitrogen) and column purified using RNeasy mini-kit (Qiagen)
Label |
Cy3
Label protocol |
Samples were labeled according to Agilent One-Color microarray-based gene expression analysis; low input Quick amp labeling protocol (v.6)
Hybridization protocol |
RNA was hybridized to Agilent bovine 4x44K slides according to the hybridization procedures in the QuickAmp Labeling kit manual. Hybridized slides were also washed and dried according to that protocol.
Scan protocol |
Slides were scanned on Agilent Technologies Scanner G2505C US85003609.
Description |
Bos taurus x Bos indicus F2 Biological replicate 21 of 47: high v low residual shear force
Data processing |
Agilent GeneSpring software (v 11.0.02) embedded functions were used for data normalization (quantile) and intepretation. Normalized to baseline median expression across all arrays, minimum threshold 1.0.
