Extraction protocol |
Total RNA was extracted with TRIzol reagent (Invitrogen) and further purified using a RNeasy Midi kit (Qiagen). DNA was removed by DNase digestion with the RNase-Free DNase set (Takara Bio). RNA quality and concentration was determined by analysis with an Agilent 2100 bioanalyzer.

Description |
This sample is of a rpoH1 rpoH2 double mutant of Sinorhizobium meliloti 1021. It is the second of four biological replicates of mutant cells with heat shock for 10 min, each from separate cultures.

Data processing |
The raw data (.pair file) was subjected to RMA (Robust Multi-Array Analysis; Irizarry et al. Biostatistics 4(2):249), quantile normalization (Bolstad et al. Bioinformatics 19(2):185), and background correction as implemented in the NimbleScan software package (Roche NimbleGen, Inc.).
