Treatment protocol Cells in collagen coated 24 well plates (BD, Franklin Lakes, NJ) were treated with 3 concentrations of chemical or DMSO vehicle for 6 h.
Growth protocol HepaRG human hepatoma cells were purchased from Biopredic International (Rennes, France). Differentiated HepaRG cells were grown following suppliers’ protocol (Biopredic) in Williams E medium (Invitrogen), supplemented with 2mM Glutamax (Invitrogen), 10% ADD670 supplement (Biopredic International) and maintained at 37°C in an atmosphere of 5% CO2.
Extracted molecule total RNA
Extraction protocol Cells were extracted in QIAgen RLT buffer and total RNA was extracted using the Agencourt RNAdvance Tissue protocol implemented on a Beckman FXp robot using methods provided by Beckman Coulter.
Label biotin
Label protocol 500 ng of total RNA was converted to Affymetrix GeneChip targets using the Affymetrix IVT-Express kit and protocols provided by Affymetrix.
 
Hybridization protocol GeneChip targets were hybridized on Affymetrix U219-96 plates using the GeneTitan single-channel instrument and protocols provided by Affymetrix.
Scan protocol GeneTitan plates were scanned on a GeneTitan single-channel instrument using protocols provided by Affymetrix.
Description Chemical signature/dose response study
Data processing Data was processed using the PLIER algorithm implemented from the Affymetrix Command Console Software.
