Treatment protocol Embryos and tissues were taken from various stages under the dissecting scope. Selected hearts were rinsed with ice cool PBS and placed in the Trizol solution (GibcoBRL).

Extraction protocol Cardiac myocyte RNA was isolated using TRIZOL®Reagent (Sigma). RNA quality was assessed using an Agilent 2100 bioanalyzer.
Label Biotin Streptavidin-Cy3
Label protocol Biotinylated cRNA were synthesized by the Ambion total prep RNA amplification kit. cDNA was quantified and normalized to 77 ng/mcL
 
Hybridization protocol Total RNA (100 ng) was hybridized to the Sentrix® MouseRef-8 Expression BeadChip (Illumina, Inc. San Diego, CA) that contains probes for ~20,000 transcripts
Scan protocol GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
Description Gene expression data from embryos younger than nuclear cycle 9, i.e. before zygotic genome activation.
Data processing The data were analyzed with Illumina Inc. BeadStudio version 1.5.0.34 and normalized by rank invariant method.
