Treatment protocol |
Mice were sensitized oropharyngeally with 100 ug ovalbumin (OVA)/ 1250 ng standard flagellin (stFLA) for 2 hours in vivo. After this time, lungs were isolated and digested with elastase and DNase to generate a single cell suspension. Cells were stained for flow cytometry and sorted for 7-AAD- CD31- CD34- CD45- EpCAM+ cells to include AECs but not endothelial cells or leukocytes.

Extraction protocol |
RNA from sorted AECs was isolated using the Trizol extraction protocol. RNA was concentrated and excess organic solvents removed using the Qiagen Rneasy MinElute Cleanup Kit.
