Treatment protocol Mice were sensitized oropharyngeally with 100 ug ovalbumin (OVA)/ 1250 ng standard flagellin (stFLA) for 6 hours in vivo. After this time, lungs were isolated and digested with collagenease, DNase, hyaluronidase, and Liberase TM to generate a single cell suspension. Cells were stained for flow cytometry and sorted for autofluorescence hi and CD11c hi cells to include alveolar macrophages but not cDCs, moDCs, or inflammatory DCs.
Extracted molecule total RNA
Extraction protocol RNA from sorted alveolar macrophages was isolated using the Trizol extraction protocol. RNA was concentrated and excess organic solvents removed using the Qiagen Rneasy MinElute Cleanup Kit.
Label biotin
Label protocol 50 ng of each total RNA sample was labeled according to the manufacturer's instructions.
 
Hybridization protocol Hybridization was done according to the manufacturer's instructions.
Scan protocol Gene expression was quantified on the nCounter Digital AnalyzerTM.
