Double-mycorrhized P. sylvestris seedlings were produced using C. geophilum 1.58 and the abundant pine symbiotic ECM species Rhizopogon roseolus (Corda) Th. Fr. or Suillus granulatus (L.) Roussel. Four months after inoculation, three and two replicate plants were harvested for each fungal combination and for non-mycorrhizal plants, respectively. Single fine root tips were collected for each ECM species per plant and for non-mycorrhizal plants in separate tubes and were shock-frozen.

Total RNAs were extracted using the NucleoSpin RNA Plant kit (MACHEREY-NAGEL) or the Power Plant RNA isolation kit (MOBIO), and cDNA was amplified using the SMART PCR cDNA Synthesis Kit (Clontech) according to the manufacturer's instructions.
