Treatment protocol |
Tibialis anterior muscles from C57Bl/6 mice were each injured with 30 µL of 50% v/v glycerol and injected with the indicated vivo morpholino (VMO) after 3 days. FAPs were then isolated from the muscles 2 days post-VMO injection.

Growth protocol |
RNA was isolated from cells that were pelleted and snap-frozen immediately after isolation from the muscle.

Total RNA was extracted using the RNeasy Mini Kit as per the manufacturer’s instructions (Qiagen).

Biotinylated single stranded cDNA were prepared according to the standard Affymetrix protocol from 100 ng total RNA.

Fragmented biotin-labeled single stranded DNA was hybridized to Affymetrix Mouse Gene 1.0 Array according to the standard Affymetrix protocol. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G.

Gene expression data from WT fibro/adipogenic progenitors isolated from WT C57Bl/6 mice injured with glycerol, injected with Control VMO 3 days post-injury, and subsequently isolated 2 days post-VMO injection.

Data processing |
The data were analyzed with the Affymertix Expression Console software using Affymetrix default RMA analysis settings.
