Title |
IRE1_L4440_1 |
Sample type |
RNA |
Source name |
Larval stage L1 to young adult, RNAi
Organism |
Caenorhabditis elegans |
Characteristics |
genotype/variation: IRE1 knockout developmental stage: young adult RNAi: pL4440 (Empty vector)
Treatment protocol |
Worm growth medium contains 25µg/mL carbenicillin and 1mM IPTG, grown from synchronised L1 stage to L4/young adult stage) 25µg/mL carbenicillin, 1mM IPTG, 25µg/mL Tunicamycin
Growth protocol |
Synchronised C. elegans at L1 stage were grown on normal growth media with 25µg/mL carbenicillin and 1mM IPTG and harvested when most of the animals had reached young adulthood.
Extracted molecule |
total RNA |
Extraction protocol |
RNA was extracted with Trizol and purified with QIAGEN RNeasy Columns
Label |
biotin
Label protocol |
Biotinylated cDNA was prepared using the Affymetrix WT PLUS Reagent Kit from 300ng total RNA
Hybridization protocol |
3 ug of fragmented and labeled cDNA were hybridized for 16 hrs at 45C on GeneChip C elegans ST arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450
Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G
Data processing |
Data was processed using RMA Normalized differential gene expression files on Series record The resulting CHP files are generated by the Affymetrix Tiling Analysis Software (v. 3.0), where gene expression of untreated samples are compared against the pmt-2(RNAi) treated samples.
