Title |
postnatal skeletal muscle at P1, biological rep1 |
Sample type |
RNA |
Source name |
Rat newborn skeletal muscle, first day of life
Organism |
Rattus norvegicus |
Characteristics |
strain: Wistar rat age: 1-day old newborn tissue: skeletal muscle
Treatment protocol |
The fetuses were delivered by Caesarean section after sacrifice of pregnant Wistar rat mothers. Adult P90 controls were collected. All tissues were snap frozen in liquid nitrogen immediately. Tissues were stored for further analyses at -80 °C.
Growth protocol |
The animals were kept and all rat tissues were obtained in the Department of Genetics of Model Diseases, Institute of Physiology CAS (Prague, Czech Republic). The experiments were performed in agreement with the Animal Protection Law of the Czech Republic and were approved by the Ethics Committee of the Institute of Physiology AS CR (Prague, Czech Republic).
Extracted molecule |
total RNA |
Extraction protocol |
TriReagent extraction of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5.5 ug total RNA (RIN>8) using Ambion WT kit.
Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45 °C on Rat Gene 1.0 ST Array. Arrays were scanned according to the protocol described in WT Sense Target Labeling Assay Manual (FS450_0007).
Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000.
Description |
RaGene1_120313W_JZ01_A1 RaGene1_120313W_JZ01_A1 Gene expression data from neonatal skeletal muscle tissue, first postnatal day. Postanatal adaptation proceeds.
Data processing |
The data were analyzed with Expression console, using Affymetrix default analysis settings and global scaling as normalization method.
