Treatment protocol |
Ovaries/testes were removed from euthanized animals after perfusion with PBS and snap-frozen in liquid nitrogen (no perfusion for P3). Adult expression profiles represent single organ samples (single animals), whereas P3 profiles represent pools of >10 females per chip.

Growth protocol |
Mice were housed in specific pathogen-free controlled conditions. Food and water were available ad libitum. All experiments were carried out in accordance with the principles and guidelines of the LAR/European Molecular Biology Laboratory (EMBL).

RNeasy (Qiagen) RNA isolation from animal tissues according to standard protocol, including on-column DNase digestion.

Biotinylated cRNAs were prepared according to the standard GeneChip WT Sense Target Labeling Assay Manual protocol (Affymetrix) from 300ng total RNA. For the P3 samples, 1µg RNA was labeled according to the CodeLink Bioarray Processing Protocol (Amersham).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 17hr at 45C on Affymetrix Mouse Genome 430 2.0 Arrays (P3) and Affymetrix Mouse Gene 1.0 ST Arrays (Adult). GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix Scanner 3000 7G.

Data processing |
Data were analyzed with RMA using Affymetrix default analysis settings (Expression Console). Up- and downregulated genes were identified using the GeneSpring GX statistical software package (Agilent).
