Sample GSM461979 Query DataSets for GSM461979
Status Public on Oct 20, 2009
Title PPAR WT_ctr_52
Sample type RNA

Source name liver, PPAR WT_ctr_52
Organism Mus musculus
Characteristics gender: male
age: 7.5 mo +/-2.5 weeks
genotype/variation: PPAR WT
treatment: control
Treatment protocol Wild-type and PPAR(alpha)-null mice were given four daily gavage doses of DEHP (1150 mg/kg/bw/day) in corn oil or corn oil alone.
Growth protocol Food (LabDiet 5001, PMI Nutrition International, St. Louis, MO) and municipal tap water were provided ad libitum. Animal facilities were controlled for temperature (20°C–24°C) and relative humidity (40%–60%) and kept under a 12-h light-dark cycle.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated and purified from mouse livers according to the mirVana™ miRNA isolation kit (Ambion, Austin, TX) The integrity of each RNA sample was determined using an Agilent 2100 Bioanalyzer (Agilent, Foster City, CA), and RNA quantity was determined using a Nanodrop ND-1000 (Thermo Fisher Scientific, Wilmington, DE).
Label biotin
Label protocol Total RNA (300 ng per sample) was labeled using the Affymetrix GeneChip WT cDNA Synthesis and Amplification kit protocol.

Hybridization protocol following Affymetrix protocol
Scan protocol The fluorescence intensities were measured with Affymetrix GeneChip scanner 3000 with 7G upgrade
Description PPAR(alpha)-null mice (129S4/SvJae-Pparatm1Gonz/J, stock #003580) and wild-type mice (129S1/SvlmJ, stock #002448) were originally purchased from The Jackson Laboratory (Bar Harbor, ME) and maintained as an inbred colony on the 129/Sv background
gene expression data from wildtype mouse liver
Data processing The intensity data files (.cel files) were loaded to Rosetta Resolver (Rosetta Biosoftware, Seattle, WA) for analysis.
