single oocyte or their surrounding cumulus cells pool were lysed using cell lysis buffer [10 U RNasein plus (Promega), 10% RealTime ready Cell Lysis Buffer (Roche), 0.3% NP40 (Thermo Fisher Scientific), and RNase-free water (TAKARA)] in 0.2 mL single tube. RamDA-seq sequencing libraries were constructed by using the advanced method of RamDA-seq (refer to the Methods section in "Single-cell full-length total RNA sequencing uncovers dynamics of non-polyadenylated RNAs, recursive splicing and enhancer RNAs" DOI: https://doi.org/10.1038/s41467-018-02866-0). However, to adapt to the manually picked single oocyte and their cumulus cells, the reaction volume from the cell lysis to the genomic DNA digestion step was modified.
