bone marrow stem cells were harvested from the femurs of C57Bl/6j mice and grown in RPMI suplemented with 10% FBS, 50 μM 2-βMercaptoethanol, 5 ng / mL murine GMCSF and 200 ng / mL murine FLT ligand. Medium was changed at 4 days and suspension cells were replated into fresh plates at 9 days. 16 days post isolation DCs were used for downstream experiments.
