Treatment protocol The exponentially growing cells were irradiated with 2 Gy of γ-radiation using a 4 MV linear accelerator (Clinac 4/100, Varian, Palo Alto, CA). No irradiated cells were used as a negative control. The irradiated and control cells were incubated for various periods of times (0, 2, 4, 8, 12 and 24 hours) before isolating total RNA.
Growth protocol The lung cancer cell lines H460 and H1299 were maintained as a monolayer in RPMI 1640 medium supplemented with 10 % fetal bovine serum, 100 units/ml penicillin, 100 μg/ml streptomycin and 2 mM L-glutamine at 37°C in humidified 5% CO2 .
Extracted molecule total RNA
Extraction protocol mirVana miRNA Isolation Kit(Ambion) extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Fragmented and Labeled DNA Target were prepared according to the standard Affymetrix protocol from 100ng total RNA (Whole Transcript (WT) Sense Target Labeling Assay Technical Manual, 2009, Affymetrix).
 
Hybridization protocol Following fragmentation, 5.5 ug of labeled DNA were hybridized for 16 hr at 45C on GeneChip Mouse Gene 1.0 ST Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the Affymetrix GeneArray Scanner GCS3000 7G plus.
Description Gene expression data from H460 4hr after ionizing irradiation (2 Gy)
Data processing The data were analyzed with Bioconductor in R statistical software.
