cDNA libraries were induced by growing S. cerevisiae in media containing galactose (YPGR) or uninduced by growing in media containing glucose (YPD).

All cells were grown in the indicated liquid media. E. coli were grown at 37˚C and S. cerevisiae were grown at 30˚C. G418 was added to all S. cerevisiae cultures to maintain plasmid selection.

mRNA and plasmid extracts were all performed using standard techniques.
mRNA was isolated with Oligo(dT)25 Dynabeads (Ambion) and libraries were preparerd with the NEXTFLEX Rapid Directional RNA-seq Kit (PerkinElmer). cDNA sequencing libraries were prepared with the NEXTFLEX Rapid DNA-seq Kit (PerkinElmer).
